1. Solid medium

After the specimen or liquid culture is streaked onto the surface of the solid medium, the individual bacteria are divided and propagated to form a group of bacteria visible to the naked eye, called colony.

(1) Morphological characteristics of colonies:
Size, shape (dew-drop, round, cauliflower, irregular, etc.), protrusions or flat, concave, edges (smooth, wavy, jagged, curled, etc.), color (red, off-white, black, green, none Color, yellow, etc.), surface (smooth, rough, etc.), transparency (opaque, translucent, transparent, etc.) and viscosity. According to the different surface characteristics of bacterial colonies, colonies can be divided into 3 types:

1 Smooth colonies (S-type colonies): The surface of the colonies is smooth, moist, and the edges are neat, and the newly isolated bacteria are mostly smooth colonies.
2 Rough colonies (R-type colonies): The surface of the colonies is rough, dry, wrinkled or granulated, and the edges are mostly irregular. Most of the R-type colonies are S-type bacterial mutations that lose the surface polysaccharide or protein formation. The R-type bacterial antigen is incomplete, and the virulence and anti-phagocytic ability are weaker than the S-type bacteria. However, there are also a few newly isolated virulence strains of bacteria, such as B. anthracis, Mycobacterium tuberculosis, and the like.
3 Mucus-type colonies (M-type colonies): The colonies are thick, shiny, and watery. More common in thick capsules or rich mucus layer of bacteria, Mycobacterium tuberculosis and so on.

(2) Colony hemolysis characteristics:
There are three cases of colony hemolysis:
1α hemolysis: also known as grass green hemolysis, 1~2mm grass green ring appears in the medium surrounding the colony, which is caused by methemoglobin;
2β hemolysis: also known as complete hemolysis, forming a completely clear and transparent hemolysis ring around the colony, which is caused by the complete hemolysis of the hemolysin produced by the bacteria;
3γ hemolysis: that is, it is not hemolyzed, the medium surrounding the colony does not change, and the red blood cells are not dissolved or defective.

(3) Pigment: Some bacteria produce water-soluble pigments, so that the colonies and the surrounding medium appear green, golden yellow, white, orange, lemon color, etc., and the resulting pigments are water-soluble or fat-soluble.

(4) Odor: Some bacteria can produce special odor after growth and reproduction in the medium, such as Pseudomonas aeruginosa (ginger smell), Proteus (chocolate charred smell), Anaerobic Clostridium (corrupted evil) Odor), Candida albicans (yeast flavor) and actinomycetes (soily taste).

2. There are three kinds of growth phenomena in liquid culture medium: most bacteria are uniformly turbid after growth and reproduction in liquid medium; a few chain-like bacteria such as streptococci and Bacillus anthracis are precipitated and grow; Obligate aerobic bacteria such as bacilli, Mycobacterium tuberculosis, and Pseudomonas aeruginosa generally grow on the surface, often forming a membrane.

3. The semi-solid medium semi-solid medium is mainly used for the bacterial dynamic test. In addition to the growth of the flagellated bacteria along the puncture line, feathery or cloud-like turbid growth can also be seen on both sides of the puncture line. The flagella-free bacteria can only grow linearly along the puncture line, and the medium on both sides of the puncture line is still clear and transparent, which is negative for the dynamic test.
