Rabies Vaccine (Vero Cell) for Human Use, Freeze-dried is a preparation of rabies fixed virus CTN-1V inoculated into Vero cell. After cultivation by bioreactor, harvest the virus suspension; further perform the concentration, inactivation, and purification. Oviposition method (1) Dry and watery stimulation method: Dry 2 to 6 hours, then drain and spawn 2 hours after running water.
(2) Dry, flowing water, and ammonia added seawater: dry 2 to 6 hours, and after 2 to 6 hours of running water, add 0.25 ton of ammonia in sea water to spawn and ejaculate.
(3) Dry, still water, and ammonia seawater: dried for about 4 hours in the shade, 2 hours in still water, and then drained in 0.25 ‰ ammonia water.
(4) Dry and ammonia-added seawater induction: After drying for 2 to 4 hours, 0.2 to 0.3 ‰ of ammonia in seawater is added.
Among them, three (2), (3), and (4) were immediately discharged into the new pool after being discharged from the parent bees, and the resulting eggs and sperm were not affected by the ammonia seawater on the fertilized eggs.
The above four methods can induce pro-Bet emission of sperm and eggs, in which the maturation effect of maturation progenitor dried and post-flow ammonia water is better.
According to experience, pro-shellworms are better at inducing oviposition after a period of cultivation in the prosperous breeding period, and their induction rate can reach 60 to 80%.
Washing eggs and hatching (1) Washing eggs: When pro-shellfish discharge, it is very easy to cause too much sperm, affect the hatching of embryos and deteriorate the water quality of nursery, generally need to wash eggs 2 or 3 times to improve the hatching rate.
(2) Hatching: The time required for embryonic development is shortened with increasing temperature. At 15-30°C, the incubation time is 60-10 hours, but the hatching rate decreases as the water temperature rises, generally at 20°C. It's better. After the D larvae are reached, they are selected for breeding.
Larvae cultivation (1) Suitable conditions for larval rearing are as follows: salinity of seawater is 13‰ to 33‰; water temperature is 18~23°C and it grows faster and develops better.
(2) Changing the water: The larvae that have been bred are only added water in the first three days, and they are constantly stirred or inflated, and then the water is changed every day.
(3) Feeds: Isochrylica, Phaeodactylum tricornutum, and Chaetoceros muelleri are fed daily in amounts of 10,000 to 60,000 cells/ml. It is fed 2 to 4 times and increases with the development of larvae.
(4) Attachment base: When the shell is as long as 130 to 145 μm, the larvae enters the shell top and metamorphosed at the later stage. Yellow sand is used as the bottom material, and the yellow sand is fully cleaned and disinfected beforehand. At 250 to 270 μm, the face plate has disappeared, the seedlings have used full movement, and the outlet pipe has formed.
(5) Cultivation of seedlings: When the water is changed, it is necessary to suck out the dirt on the surface of the bottom sediment to prevent the pollution of the bottom matter; after changing the water every day, feed the mixture of Platycodon and Chrysophyta. When it reaches 1.2mm, the inlet pipe is formed. Its length is about half of the outlet pipe, reaching 0.5cm can be out of the tank for intermediate cultivation.
